Connecticut!

Connecticut is very interesting. They use a simple white box with black numbering and outline, but when seen on green navigation signs, the white turns green, and the black turns white.

Regarding Connecticut, I can't help but notice that the U.S. route in that symbol seems to function the same way: it becomes a white outline that's filled in with green. That's pretty interesting. But I went to look up further examples, and that doesn't seem to consistently happen?
Situations where it does: CT-25/US-1, CT-15/US-7
Situations where it does not: US-5/I-91, US-1/I-95, US-202/US-7

I assumed it was something like the Oregon Street Limit Signs where it is restricted on Interstates (though seemingly not city limits in this case), but then I came across this, which has the normal coloration for both U.S. and State routes... on a state route. So I don't even know anymore.

I agree, CT is all over the place with signage. The overhead signs and roadside navigation signs use the green/white combo like you posted, but also a black/white version. It's possible that the mix is because they're changing signs to a different standard, and luckily there's going to be some time before SCS gets there. The only consistent signs I saw were the standalone signs. Unlike a lot of other states, the shape of the standalone signs is always a square. However, the shape of 3 digit overhead and roadside navigation signs are rectangular.

Here is an older pic of an interchange where the closest sign uses the green/white scheme. On the streetview though, the sign that's further down the road uses the white/black scheme.

Based solely off the examples here, it seems that not colouring in the US route shield is an outdated practice. If you notice, in all of the examples where the shield is not colored in, the sign has other indications that it is old such as the exit tab being in the centre of the sign and the reflective material on the sign is worn. On the signs where the US route shield is coloured in, the sign has updated designs and better visabilty/reflectivity. To know for sure though, we would need to consult the most recent version of the Connecticut MUTCD supplement.

Rhode Island
Rhode Island uses a simple white square with a black border for 1 and 2 digit routes, and a white rectangle with a black border for 3 digit routes. All signs have either "R.I." or "RI" above the route number, and the sign design doesn't change between navigation and standalone signs.

Massachusetts
1 and 2 digit routes use white square signs with black borders, and 3 digit routes are rectangles. Most overhead and roadside navigation signs have no black border, but there are exceptions.

Pretty much the same as Massachusetts (white with black borders), except all the overhead and roadside signs omit the black border. 1 and 2 digit routes are square signs, 3 digit routes are rectangles.

Vermont
Vermont's signs are white with green backgrounds. "Vermont" is in a separate rectangle with rounded corners above the route number. The route numbers are contained in a flattened oval shape. 1 and 2 digit routes are square signs, 3 digit routes are rectangles. The color scheme does not change between the standalone and the navigation signs, but the navigation signs have an additional white outline.

New Hampshire
The shield for New Hampshire uses the silhouette of the (now collapsed) "OId Man of the Mountain" rock formation. On standalone signs the background and outline is black; on navigation signs only the white silhouette is used. There is no difference in the width or height of 1, 2 or 3 digits signs. Note the different width of the number characters between 2 and 3 digit signs.
