As a transmission guy, I can say some things about number of gears. Actually, you will barely ever need more than 20 gears, because they can give you all the needed range and a little step between the gears at the same time. The second thing is, 10-speeders were never supposed to be paired with auxiliary transmissions. They were made to be good on their own. Well, you can try pairing it with a 2-speed box and see what you get. But I've barely seen anything like that IRL.

Ever since 1.50 came out, I haven’t seen the transmissions in your mod be available to select when building trucks. Does it need an update, or is there something screwy with the latest update of the game?

Well, I guess that's because they are applied for Peterbilt 389 only, but they should be done for the trucks you want yourself (described in the main post). And yes, it should work for 1.50 as is.
Actually not that I'm currently supporting this mod... For a reason that SCS does not let you change final gear ratio without modifying the files, and also no support of torque limit for each gearbox (unless you manually define which exact engines are supported, but that won't work for modded engines for example). Given this, I make custom gearboxes for myself only. But I always can give you an advice

I have created additional folders for the trucks I want those transmission to be used in, but I don’t see the transmissions as options in those trucks anymore when building them - since the update. They have worked in the past, but they don’t show up anymore. I have also changed the 3rd line of each transmission file to reflect the def name of the truck. Weird.

I'm trying to set up my own transmission mod, using 1 transmission as a test. I think I have it set up right (based on your mod structure as a template, since I'm new to turning my stuff into a mod), but the transmission isn't showing up as an option in the truck (Harven's Mack R). Here is a link to what I have put together. Any advice would be great. I understand this is beyond the scope of what you're doing with the Spicer DIY mod. It all looks nice, but the folder name is not "transmissions" but "transmission". I'm mostly sure that's the reason.

I know nothing about Spicer transmissions, but want to try this mod out, Hope you don't mind me asking some questions:
How many gears was supported on a single stick, as I don't have 2 sticks to use ?
Do you have any good reference material for Spicer that I can read ?
Are there any ready made shifter layouts floating around, alternatively an understandable diagram I can use to modify the default ones ?

Hello!
1) It differs from truck to truck and of course game has no limitations for that - I was driving some of the old trucks with 5 or even 4 gears in total as a manual transmission. Six or seven gears seems the best option as a wide-range gearbox. And the 10-speed one is good too with only one range splitter if I remember it correctly. Actually you can simulate a 6x4 gearbox too if you replace the second stick with both range splitters (the same way as the 18-speeder works in the game already).
2) Frankly speaking I do not own (or know a way to find) any references apart from different specification sheets.. Because they mostly have all the info I need to simulate the transmission in the game, sometimes including max torque, RPM and shift patterns.
3) I'd recommend trying making all the needed layouts yourself, as it's individual for each gearbox. I hope there is a guide how to make them, but I got to knowing it myself somehow. You just need to program each position of your gearbox, like.. pointing the "3" physical position means the 5th gear in the game in low range and 10th in high range. And making full list of these combinations. Given this, you can simulate any real gearbox and even modify the template if you wish so (for example I could make ZF 9-speeder without range splitter at all).
